Effect of storage time and effect of sonication on decellularization of porcine esophagus

Esophageal disorders require surgical reconstruction using either alternate substitutes or scaffolds. Tissue engineering have long been touted to be one of the ways to replace diseased or defective organs. This research uses tissue engineering to obtain decellularized esophagus scaffolds. By obtaining decellularized esophageal tissues, we can accurately maintain the extracellular matrix which provides a suitable microenvironment for recellularization. The aim of this study is to determine if storage time affects decellularization and also if ultrasonic affects decellularization of porcine esophagus. Three set of experiment were done using sample that were kept for 0 days, 11 days and 33 days respectively. The experiment is conducted by soaking porcine esophagus in 1% sodium dodecyl sulfate (SDS) and put into ultrasonic machine which is set to run for 15 minutes and rest for 30 minutes. Histology showed that there is no difference between the samples from different storage time after 3 days, 5 days or 7 days of decellularization done. A control sample of 11 day old esophagus is soaked in 1% SDS and used with a shaker to compare if ultrasonic machine helps decellularization. Histology shows that ultrasonic helps to remove the nuclei from the muscle area faster. We can see this especially after 3 days of decellularization done. For shaking, there are still small amount of nuclei present while for ultrasonic, there is no nuclei present in the muscles area. Therefore, we can conclude that storage time does not affect decellularization and ultrasonic helps decellularization but only confine to the muscle area of the esophagus.
